Marvel has had gay characters since the late 90's. Northstar being the most notible of their characters at the time.
DC has been a little slower in bringing gay characters into their stories. Of course I am a little skeptical about a "main character" turning out to be gay. I really don't expect Superman, Batman, or any of their big names to be changed to such a degree.

If you read the older issues, you can also see a ton of parallels between the comics and the 19th century civil rights movement, with Xavier and Magneto being based upon Martin Luther King and Malcolm X, respectively.

Well, it's been said it will be a "major, iconic character". It's also been said to be a male character.
That basically boils it down to Superman, Batman, Flash, or Green Lantern. It's be really hard to classify anyone else as both "major" and "iconic". Though with Green Lantern, they do have like four different ones they could pick from.
Plus, after all the Batman and Robin jokes, it would just be tacky.
In all honesty, if it's one of the big ones, it's Wonder Woman. While a ret-conned gay Flash or Batman or Supes might really upset the fanboys, a lesbian WW would send them into fits of nerdgasmic ecstasy.In their defense Booster was the star of the well received 52
But, as someone else mentioned, DCs idea of "iconic" and "major" is pretty damn broad. For God's sake, they consider Power Girl, Booster Gold, and Beast Boy to fit the definition.
I would say the most likely candidates:
Power Girl
Cyborg
Some B list hero from either Stormwatch or the Justice Society

Martian Manhunter's been a part of the Justice League cartoons for the past decade. I'm not terribly familiar with comics, but I think that makes him a bit more than a minor character.
Plus, is anyone else amused that they chose the character whose official vulnerability is "wood" to be gay...?
